The Cromolyn-binding protein (CBP), also known as the mast cell membrane Ca2+-binding protein, is a 78 kDa membrane-associated protein that serves as a critical regulator of calcium signaling in mast cells and basophils (Mazurek et al., 1984). It acts as a gatekeeper for the influx of extracellular calcium ions, a process that is essential for the degranulation and release of inflammatory mediators such as histamine, leukotrienes, and cytokines (Hemmerich & Pecht, 1991). When mast cells are triggered by IgE-crosslinking, this protein facilitates the opening of calcium channels; however, the binding of mast cell stabilizers like cromolyn sodium (disodium cromoglycate) to CBP prevents this activation (PubChem, CID 2850). This stabilization of the mast cell membrane effectively inhibits the allergic cascade and prevents the symptoms of Type I hypersensitivity. The protein is a primary therapeutic target for managing conditions such as bronchial asthma, allergic rhinitis, and systemic mastocytosis (DrugBank, DB01003). While its precise molecular identity has been historically elusive and sometimes associated with proteins like S100A13 or nucleoside diphosphate kinase, it remains a foundational concept in the pharmacology of anti-allergic medications.
