Crooked neck pre-mRNA splicing factor 1 (CRNKL1) is a ubiquitously expressed nuclear protein and a component of the spliceosome, essential for pre-mRNA splicing, particularly for U12-type introns. It functions as a nuclear retention factor to regulate the export of intron-retaining RNAs and is involved in the processing of HIV-1 unspliced RNAs, helping to control the cytoplasmic levels of both viral and cellular intron-containing transcripts. Loss or depletion of CRNKL1 leads to an increase in cytoplasmic levels of unspliced RNAs and influences genes involved in transcription, the cell cycle, and organelle organization. It has been associated with Fanconi anemia and may play roles in cancer and other diseases linked to splicing dysregulation[1][2][3][5][8].
