Cross-Reactive Material 197 (CRM197) is a non-toxic mutant of diphtheria toxin used as a carrier protein in conjugate vaccines. The mutation (G52E) abolishes ADP-ribosyltransferase activity, eliminating toxicity, while preserving the protein's structure and lysine residues for conjugation. It enhances the immunogenicity of polysaccharides and haptens, eliciting stronger immune responses. CRM197 is also explored as an HB-EGF inhibitor and for targeted drug delivery.
Acts as a carrier to present weakly immunogenic antigens to the immune system, facilitating T-cell help and enhanced antibody production. Can also inhibit HB-EGF signaling.
