Overview

Crumbs cell polarity complex component 2 (CRB2) is an evolutionarily conserved transmembrane protein involved in the formation and maintenance of epithelial and neuronal cell polarity, as well as structural organization in tissues such as the retina and kidney. CRB2 is a core member of the Crumbs polarity complex, partnering with PALS1 and PATJ to determine the apical plasma membrane domain, promote tight junction formation, and orchestrate correct cell-cell adhesion in tissues. Loss or mutation of CRB2 disrupts these processes, leading to defects in tissue architecture, retinal degeneration, and congenital kidney disorders. CRB2, together with other Crumbs complex components (such as CRB1 and CRB3), is essential in specifying epithelial cell polarity and maintaining the apical membrane domain. In humans, CRB2 is expressed in important tissues including the retina, kidney, and specific regions of the brain; in the retina, it is crucial for neuroepithelium and photoreceptor morphogenesis and organization. Mutations in the CRB2 gene are associated with inherited retinal dystrophies and syndromes involving cystic kidney and brain malformations. The canonical abbreviation CRB2 is widely used in the literature. There is no evidence that CRB2 is currently a direct drug target (i.e., receptor, enzyme, or channel); its significance lies in developmental biology and disease pathology rather than pharmacological modulation. There are no described drugs, mechanisms of drug action, or validated biomarkers tied to CRB2 modulation at present.
