Cruzipain is a major cysteine protease and sulfated glycoprotein produced by the protozoan parasite *Trypanosoma cruzi*, the causative agent of Chagas disease. It is expressed in all developmental stages and strains of the parasite, playing essential roles in parasite virulence, host cell invasion, immune evasion, and tissue degradation. Due to its essential role for *T. cruzi* survival and pathogenesis—and absence from humans—cruzipain has been validated as an attractive target for drug development against Chagas disease. Multiple classes of cruzipain inhibitors have shown efficacy against *T. cruzi* both in vitro and in animal models.
