Overview

Cryptic family protein 1B (CFC1B) belongs to the EGF-CFC (Cripto/FRL-1/Cryptic) family of signaling proteins. It is a GPI-anchored co-receptor with a cysteine-rich EGF-like domain, essential for patterning the left-right axis during embryonic development. CFC1B interacts with the Nodal and Activin type I receptor ALK4, contributing to circulation, organ morphogenesis, and stem cell fate specification. It is overexpressed in multiple cancers, promotes epithelial-to-mesenchymal transition (EMT), and has been linked to poor survival and tumor aggressiveness. Mutations or dysregulation are associated with congenital disorders such as visceral heterotaxy and heart defects.
