Overview

GP25-200 is a glycoprotein complex found in the apical complex and on the surface pellicle of Cryptosporidium parvum sporozoites and merozoites [1, 3]. It consists of multiple protein species ranging from 25 to 200 kDa and is essential for the parasite's ability to attach to and invade host intestinal epithelial cells [1, 8]. Due to its critical role in the infection process and its high conservation across different Cryptosporidium isolates, GP25-200 is a major target for passive immunotherapy and vaccine development [1, 11]. Monoclonal antibodies such as 3H2 and 4H9 specifically target this complex to neutralize parasite infectivity, often showing enhanced efficacy when used in polyvalent formulations with antibodies against other antigens like CSL and P23 [1, 8]. Additionally, GP25-200 is being utilized as a targeting moiety for nanoparticle-based drug delivery systems to improve the treatment of cryptosporidiosis, particularly in immunocompromised individuals where current therapies are often inadequate [6, 10].

Mechanism of action

Neutralization of parasite infectivity by inhibiting attachment to and invasion of host intestinal epithelial cells; induction of circumsporozoite precipitate (CSP)-like reactions; and targeted delivery of biocides or drug-loaded nanoparticles to the parasite surface [1, 6, 10].
