Overview

CTD small phosphatase-like protein 2 (CTDSPL2) is an enzyme that localizes primarily in the nucleus and acts as a serine/threonine phosphatase for the C-terminal domain of RNA polymerase II[6][2][3][1]. It plays a crucial role in regulating transcription, protein dephosphorylation, and signaling pathways such as PI3K/AKT via JAK1 upregulation[1]. CTDSPL2 is involved in glucose metabolism and can promote hepatic gluconeogenesis and muscle proteolysis[1]. In various malignancies, particularly non-small cell lung cancer and acute myeloid leukemia, CTDSPL2 overexpression promotes proliferation, migration, invasion, and metastasis of cancer cells. It can also inhibit immune cell (CD4+ T cell) infiltration within tumors, supporting tumor immune evasion[1]. CTDSPL2 is a direct target of the tumor suppressor microRNA miR-193a-3p, which negatively regulates its expression. Elevated levels of CTDSPL2 are linked to poor prognosis in certain cancers, highlighting its potential as a therapeutic target and biomarker[1][2][6].
