CTTNBP2 N-terminal-like protein (CTTNBP2NL) is a paralog of cortactin-binding protein 2 (CTTNBP2) and is highly conserved across mammalian species[1][4]. It shares moderate sequence similarity, especially at the N-terminus and coiled-coil domain, with CTTNBP2, but its expression profile and functional roles are distinct. Unlike CTTNBP2, which is predominately expressed in the brain and regulates dendritic spine formation and synaptic protein distribution, CTTNBP2NL is expressed at low levels in the brain and instead localizes to actin stress fibers, not to neuronal dendritic spines[1][4]. CTTNBP2NL interacts with cortactin and striatin family proteins, suggesting a role in cytoskeletal arrangement and potentially in PP2A regulatory complexes[1]. However, current evidence does not support a direct role for CTTNBP2NL in dendritic spine development, synaptic function, or as a disease biomarker or therapeutic target[1][4]. No drugs are known to interact with this protein, and there are no established safety concerns or mechanisms of drug action.
