Overview

CUB and sushi domain-containing protein 2 (CSMD2) is a large, single-pass transmembrane protein characterized by multiple extracellular CUB and Sushi (CCP) domains[1][2]. It is highly expressed in the brain, particularly in neurons of the neocortex and hippocampus, and localizes predominantly to dendrites and dendritic spines where it interacts with synaptic scaffold proteins such as PSD-95[1]. Functional studies demonstrate that CSMD2 is required for dendrite arborization and maintenance, as well as dendritic spine density and development, pointing to key roles in synaptic formation and function[1]. Genetic variants and altered expression of CSMD2 have been linked to neuropsychiatric diseases including schizophrenia and autism spectrum disorders[2]. In oncology, CSMD2 is upregulated in various cancers and may participate in immune modulation, complement cascade regulation, and cell adhesion; high CSMD2 expression correlates with poor prognosis and may serve as a prognostic or diagnostic biomarker in several tumor types[2]. No approved drugs or experimental therapeutics currently target CSMD2.
