CUB and zona pellucida-like domains 1 protein (CUZD1) is a type 1 transmembrane protein characterized by two consecutive CUB domains and a zona pellucida (ZP) domain, with a C-terminal transmembrane region. CUZD1 is highly expressed in pancreatic acinar cells, where it localizes to zymogen granule membranes and is implicated in trypsinogen activation. Functional studies show that CUZD1 acts as a secretory protein, and its variants can alter secretion efficiency and contribute to disease susceptibility, notably chronic pancreatitis. In addition to its pancreas-specific expression, animal studies suggest essential roles for CUZD1 in mammary gland development and proliferation, partly via modulation of the JAK/STAT5 signaling pathway and regulation of EGF family ligands. Some evidence indicates involvement in breast cancer, where overexpression may be linked to tumorigenesis. The full physiological and pathological functions of CUZD1 remain incompletely characterized.
