Overview

CUE domain-containing protein 1 (CUEDC1) is a ubiquitin-binding protein characterized by a single CUE domain, structurally related to ubiquitin-associated (UBA) domains, which facilitates protein-protein interactions involved in the regulation of protein sorting and degradation via cellular pathways such as the proteasome and autophagy[1][2][6][7]. CUEDC1 is ubiquitously expressed in human tissues and has been implicated in the control of cancer-relevant cellular processes, including epithelial-mesenchymal transition (EMT), cell proliferation, and apoptosis, particularly in non-small cell lung cancer (NSCLC) and breast cancer[2][5][6]. Experimental evidence indicates that CUEDC1 can inhibit metastasis and cell proliferation by suppressing TGF-β/Smad signaling and by stabilizing Smurf2, a negative regulator of this pathway[2][6]. Moreover, CUEDC1 is a downstream target of the estrogen receptor alpha (ERα) and is essential for proliferation of ERα-positive breast cancer cells[5]. No approved therapeutic agents directly target CUEDC1 as of the current knowledge, and its value as a druggable target is under investigation.
