CUEDC1 antisense RNA 1 (CUEDC1-AS1), also termed lncCUEDC1, is a nuclear-localized long non-coding RNA transcribed antisense to the CUEDC1 gene. It has been experimentally shown to negatively regulate stem cell-like functions in breast cancer stem cells (BCSCs), mainly by suppression of NANOG expression. Knockdown studies demonstrate that loss of CUEDC1-AS1 increases stemness markers and enhances BCSC phenotypic traits such as mammosphere and colony formation. Its regulatory role can potentially make it a candidate for further exploration as a biomarker or a therapeutic target in cancer stem cell biology. The CUEDC1 gene itself encodes a ubiquitin-binding protein (CUE domain containing 1), which is unrelated to CUEDC1-AS1 except by genomic context. No drugs have been reported to interact with CUEDC1-AS1 directly in published therapeutic pipelines.
For knockdown studies, inhibition of CUEDC1-AS1 leads to increased expression of stemness markers such as NANOG, SOX2, and OCT4 in breast cancer cells.
