CUGBP Elav-like family member 6 (CELF6) is a member of the CELF/BRUNOL family of RNA-binding proteins characterized by having three RNA recognition motif (RRM) domains (two N-terminal, one C-terminal) and a divergent domain between the second and third RRM domains[1][2][5][7]. CELF6 is expressed most abundantly in the brain and kidney. It regulates post-transcriptional gene expression, particularly by modulating pre-mRNA alternative splicing, mRNA stability, and translation. In neuronal tissue, CELF6 binds to UGU-rich motifs in the 3’ UTRs of synaptic mRNAs, influencing synaptic protein abundance and neurological function[2][5]. Genetic disruption of CELF6 is implicated in neuropsychiatric disorders, such as autism spectrum disorder, and changes in CELF6 expression have been associated with cancer, where it can act as a tumor suppressor by stabilizing the mRNA of cell cycle regulators such as p21 and influencing pathways like p53[3][4][5]. CELF6 does not currently have known direct pharmacological ligands and is not a recognized therapeutic target for drugs in clinical use.
