Overview

Cupressus arizonica pollen allergens are a group of proteins derived from the Arizona cypress (also known as Hesperocyparis arizonica) that serve as potent environmental triggers for Type I hypersensitivity. The major allergen, Cup a 1, is a pectate lyase enzyme responsible for degrading pectin in plant cell walls to facilitate pollen tube growth and fertilization [1, 15]. These allergens are a primary cause of winter pollinosis in Mediterranean climates and the Southwestern United States, manifesting clinically as allergic rhinitis, conjunctivitis, and occasionally asthma [5, 6]. In medical practice, purified or standardized extracts of these allergens are used as the active pharmacological agents in allergen-specific immunotherapy (AIT) [7, 17]. AIT works by gradually desensitizing the patient's immune system, shifting the response from a pro-inflammatory Th2 state toward a regulatory state characterized by the induction of regulatory T cells and the production of allergen-specific IgG4 blocking antibodies [3, 11]. Consequently, these allergens function as both the pathological trigger of allergic disease and the specific target for immunological desensitization therapies.

Mechanism of action

Allergen-specific immunotherapy (AIT) involves the controlled administration of Cupressus arizonica pollen extracts to induce immunological tolerance. The mechanism involves shifting the immune response from a Th2-mediated allergic phenotype to a Th1/Treg-mediated regulatory phenotype, leading to the production of allergen-specific IgG4 antibodies that block IgE-allergen binding and inhibit mast cell degranulation [3, 12].
