Overview

CXXC motif containing zinc binding protein (CZIB) is a conserved, eukaryote-specific protein encoded by the gene C1orf123, located on chromosome 1p32.2 in humans. The protein is 160 amino acids in length and contains two canonical CXXC motifs, which bind a zinc ion via tetrahedral coordination, forming a distinctive zinc-binding domain. This domain structurally resembles elements found in nucleic acid-binding proteins and may support protein regulation or interaction with small molecules, as indicated by a conformational cavity adjacent to the zinc-binding site. CZIB is part of the DUF866 protein family, whose biological role remains unresolved but is hypothesized to participate in mitochondrial oxidative phosphorylation based on homology and functional studies in yeast. There are currently no established disease associations, identified interacting drugs, or known clinical biomarkers for CZIB.
