CXXC-type zinc finger protein 4 (CXXC4, also known as IDAX) is a multifunctional zinc finger protein that antagonizes the canonical Wnt/β-catenin signaling pathway by direct interaction with Dishevelled proteins, thus negatively regulating signal transduction critical for cell proliferation and differentiation. Its CXXC domain specifically binds unmethylated CpG dinucleotides, targeting it to promoter regions and CpG islands, where it participates in chromatin modification and epigenetic regulation. CXXC4 also interacts with methylcytosine dioxygenase TET2, linking it to DNA methylation dynamics. Mutation or dysregulation of CXXC4 has been implicated in malignant renal cell carcinoma and other diseases involving aberrant Wnt signaling and epigenetic control.
