Cyclic GMP-AMP synthase (cGAS) is a cytosolic enzyme that functions as a key sensor of double-stranded DNA (dsDNA) in the innate immune system. Upon binding to dsDNA in the cytoplasm, cGAS catalyzes the synthesis of cyclic GMP-AMP (cGAMP), a secondary messenger that activates downstream immune signaling pathways. Dysregulation is linked to autoimmune diseases while targeted stimulation may be beneficial in immuno-oncology. Recent studies also show direct involvement in autophagy regulation.
