Overview

Cyclic GMP phosphodiesterase (cGMP-PDE) refers to a group of enzymes that specifically catalyze the hydrolysis of cyclic guanosine monophosphate (cGMP) into 5′-GMP, thereby regulating intracellular levels of cGMP and modulating various physiological processes. These enzymes are part of the broader superfamily known as cyclic nucleotide phosphodiesterases (PDEs), which also includes enzymes that act on cAMP. They play critical roles in visual transduction, smooth muscle relaxation, and platelet aggregation. The activity and localization of different isoforms can be dynamically regulated by hormones, neurotransmitters, light, and calcium levels. Many animal-derived or synthetic inhibitors target specific isoforms, most notably PDE5 inhibitor Sildenafil for erectile dysfunction and PDE4/7/8 inhibitors like Rolipram for anti-inflammatory research.
