Overview

Cyclic guanosine monophosphate-phosphodiesterases (cGMP-PDEs) are a family of enzymes that hydrolyze cyclic guanosine monophosphate (cGMP) to 5'-GMP, thereby regulating the intracellular levels of cGMP. These enzymes play a crucial role in controlling the intensity and duration of cGMP signaling within cells. cGMP acts as an important second messenger involved in various physiological processes, including vasodilation, regulation of cardiac function, phototransduction in vision, and modulation of cell growth and apoptosis. Inhibitors targeting specific phosphodiesterase isoforms have significant therapeutic applications, such as PDE5 inhibitors (e.g., sildenafil) which are used clinically for erectile dysfunction and pulmonary hypertension due to their ability to prolong vasodilatory effects mediated by NO-cGMP signaling pathways.
