Cyclic nucleotide-gated channel alpha subunits (typically abbreviated as CNGA) are *pore-forming* proteins that assemble into tetrameric ion channels activated by the direct binding of cyclic nucleotides (mainly cGMP or cAMP)[1][3]. They are essential in the sensory transduction pathways of photoreceptor cells (vision) and olfactory sensory neurons (smell), where they convert chemical signals (levels of cAMP or cGMP) into electrical signals by mediating sodium and calcium influx[1][3][4]. Each alpha subunit spans the membrane six times (S1–S6), has a pore-forming P-loop, and a cytoplasmic cyclic nucleotide-binding domain[1][2]. These channels are critical for the electrical responses to light and odors[3]. Mutations in specific alpha subunits—such as CNGA1—can result in severe hereditary diseases, including retinitis pigmentosa, due to disruption of phototransduction in retinal rods[4]. Their pharmacology is an area of active research, with some non-specific modulators identified, but no highly specific drugs currently in widespread clinical use[5][3].
