Overview

Cyclin D-binding Myb-like transcription factor 1 (DMTF1) is a transcription factor with a cyclin D-binding domain and Myb-like repeats, functioning as a tumor suppressor largely via activation of the ARF–p53 pathway; it mediates growth arrest and apoptosis in response to oncogenic stress (notably Ras pathway activation), and plays important roles in cell cycle regulation, hematopoietic differentiation, and transcription of specific genes such as ARF and aminopeptidase N. DMTF1 is present in multiple isoforms, with the α isoform serving as a potent transcriptional activator and tumor suppressor, while β and γ isoforms can antagonize α function, sometimes promoting proliferation. Deletions, aberrant splicing, or downregulation of DMTF1 are implicated in the pathogenesis of several cancer types.
