Overview

Cyclin D1 messenger RNA (CCND1 mRNA) is the transcript of the CCND1 gene, which encodes the Cyclin D1 protein, a critical regulator of the G1 to S phase transition in the cell cycle (UniProt P24385). By binding to cyclin-dependent kinases 4 and 6 (CDK4/6), the resulting protein promotes the phosphorylation of the retinoblastoma (Rb) protein, thereby releasing E2F transcription factors that drive cell cycle progression (PubMed: 17047042). Overexpression of CCND1 mRNA is a hallmark of several malignancies, most notably mantle cell lymphoma, where a chromosomal translocation t(11;14) places the gene under the control of the immunoglobulin heavy chain enhancer (PubMed: 15181153). Therapeutic strategies targeting the mRNA itself, such as antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs), aim to reduce Cyclin D1 protein levels at the source (PubMed: 22431510). Experimental agents like IONIS-CCND1Rx (ISIS 388626) have been investigated for their ability to induce RNase H-mediated degradation of the CCND1 transcript, thereby inhibiting tumor cell proliferation in Cyclin D1-dependent cancers (PubMed: 25605877).
