Cyclin-dependent kinase 15 (CDK15), also known as PFTK2, PFTAIRE2, or ALS2CR7, is a member of the cyclin-dependent kinase subfamily of serine/threonine kinases, structurally characterized by a conserved protein kinase domain and functional dependency on cyclin binding for activation[1][2]. CDK15 is evolutionarily conserved and closely related to other CDKs, sharing a two-lobe kinase fold that is catalytically inactive without allosteric activation by a cyclin partner (notably cyclin Y for CDK15 and its homologs)[1][2]. While most well-known CDKs (such as CDK1, CDK2, CDK4, and CDK6) regulate canonical cell cycle transitions, the biological function of CDK15 is less well defined, but evidence implicates it in signal transduction and potential cell survival pathways, especially in neuronal tissues. The gene is located in a chromosomal region associated with juvenile forms of amyotrophic lateral sclerosis (ALS2 region)[1], although direct pathogenic links remain unproven. Limited reports suggest possible involvement in tumor biology as well, but CDK15 is not a major target in current cancer or neurodegeneration therapeutics. No specific drugs, mechanisms, biomarkers, or clinical safety concerns are established for CDK15 at present. Note: The canonical abbreviation (CDK15) and aliases (PFTK2, PFTAIRE2, ALS2CR7) are consistently used in major protein, gene, and pharmacological databases, and the molecule is categorized as an enzyme of the cyclin-dependent kinase family[1][2]. Most published research and review literature treat it as a potential, but not yet validated, therapeutic target with limited direct disease association.
