The Cyclin-dependent kinase 19 (CDK19)-cyclin C complex is a key component of the Mediator complex's kinase module, playing a vital role in regulating RNA polymerase II-mediated transcription (UniProt Q9BWU1). CDK19 is a paralog of CDK8, and both kinases associate with cyclin C (CCNC), MED12, and MED13 to form a regulatory subunit that modulates gene expression in response to various signaling pathways (PMID: 24161115). Unlike cell-cycle-driving CDKs, the CDK19-cyclin C complex functions primarily as a transcriptional regulator, phosphorylating the C-terminal domain of RNA polymerase II and transcription factors such as STAT1 (PMID: 27561550). In disease contexts, CDK19 is often overexpressed in several malignancies, including prostate and colorectal cancers, where it contributes to tumor progression, metastasis, and resistance to therapy (PMID: 30333230). Small-molecule inhibitors like Senexin B and Cortistatin A have been developed to target the ATP-binding site of CDK19/CDK8, showing potential in suppressing oncogenic transcriptional programs and inflammatory responses (PMID: 26190105, PMID: 27561550). Beyond oncology, the complex is implicated in inflammatory diseases and fibrosis, making it a versatile target for drug development (PMID: 32814874). Therapeutic challenges include the high structural similarity between CDK19 and CDK8, which often results in dual inhibition, and the need to minimize effects on general transcription (PMID: 24161115). Current research focuses on identifying specific biomarkers, such as phospho-STAT1 levels, to monitor the efficacy of CDK19-targeted therapies in clinical settings (PMID: 27561550).
