Cyclin-dependent kinase 5 activator 2 (CDK5R2) is a neuron-specific protein encoded by the CDK5R2 gene. Its primary role is to bind and activate cyclin-dependent kinase 5 (CDK5), forming an active kinase complex essential for the development, differentiation, migration, and survival of neurons. CDK5R2 is one of two principal CDK5 activators, the other being CDK5R1 (p35); CDK5R2 (commonly known as p39) is most relevant in nervous tissue. Unlike classical cyclins, it defines a distinct family of kinase activators and is crucial for neuronal cytoskeletal organization, controlling growth cone dynamics, and modulating cell cycle-independent processes. Disruption of CDK5R2 or its complex formation can lead to severe neurodevelopmental abnormalities, and aberrant activity is implicated in certain neurodegenerative states.
