The cyclin-dependent kinase 5 (CDK5)-p25 complex is a protein kinase complex formed by the association of CDK5, a proline-directed serine/threonine kinase, with p25, a proteolytic fragment derived from its physiological activator p35. This complex plays critical roles in neuronal function and is implicated in neurodegenerative diseases due to its aberrant activation and altered substrate specificity. Hyperactivation of the CDK5-p25 complex contributes significantly to neurodegeneration seen in disorders such as Alzheimer's disease. Prolonged activation leads to aberrant phosphorylation events causing cytoskeletal disruption, synaptic dysfunction, cell death pathways activation, and ultimately neuronal loss. Mislocalization away from membranes further disturbs normal signaling pathways.
