Cyclin-dependent kinase 6 (CDK6) messenger RNA (mRNA) is the nucleic acid template responsible for the synthesis of the CDK6 protein, a key driver of the eukaryotic cell cycle [NCBI Gene, 2024]. As a member of the serine/threonine protein kinase family, the protein product of this mRNA complexes with D-type cyclins to phosphorylate the retinoblastoma (Rb) protein, thereby triggering the transition from the G1 to the S phase [UniProt, 2024]. Overexpression or dysregulation of CDK6 mRNA is frequently observed in various cancers, including hormone receptor-positive breast cancer and hematologic malignancies like acute myeloid leukemia, where it promotes uncontrolled cellular proliferation [PubMed, 2018]. While established clinical therapies utilize small-molecule inhibitors like Palbociclib to block the kinase activity of the CDK6 protein, the mRNA itself is an emerging target for RNA-based therapeutics such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs) [PubMed, 2019]. These novel approaches aim to silence CDK6 at the transcript level, potentially bypassing resistance mechanisms associated with protein-targeted drugs and addressing the non-catalytic roles of CDK6 in tumor progression [PubMed, 2021]. Furthermore, targeting the mRNA allows for the modulation of CDK6 expression in tissues where protein-level inhibition may be insufficient or where the protein acts as a transcriptional co-regulator independent of its kinase activity [PubMed, 2020].
