Cyclin-dependent kinase 8 (CDK8) is a serine/threonine protein kinase that belongs to the cyclin-dependent kinase family. It forms a complex with cyclin C and is primarily known for its role in regulating transcription as part of the Mediator complex. CDK8 is an oncogenic driver in multiple cancers, activating pathways like Wnt/β-catenin and regulating estrogen-inducible genes. High CDK8 levels are associated with poor prognosis in certain cancers. It functions through phosphorylation events affecting target proteins involved in gene regulation. Inhibitors like Senexin A block its ATP-binding pocket.
