Cyclin-dependent kinase inhibitor 1B (CDKN1B) encodes the protein p27Kip1, a critical negative regulator of cell cycle progression. It binds to cyclin-CDK2 and cyclin-CDK4 complexes, inhibiting their kinase activity and thereby enforcing cell cycle arrest at the G1 phase. As a tumor suppressor, loss or downregulation of p27 is frequently observed in a wide range of human cancers and is associated with poor prognosis. CDKN1B is also implicated in inherited tumor syndromes, notably multiple endocrine neoplasia type 4. Therapeutically, modulation of this pathway is a key mechanism for many anti-cancer agents, especially CDK inhibitors[1][2][3][6].
