Cyclin-dependent kinase inhibitor 3 (CDKN3) is a dual-specificity protein phosphatase that negatively regulates cell cycle progression by dephosphorylating cyclin-dependent kinases such as CDK1 and CDK2, most notably at activation sites Thr-161/Thr-160. CDKN3 acts as a key cell cycle regulator and is associated with inhibition of cell proliferation; however, its function is context-dependent, and dysregulation or overexpression in tumor tissues leads to enhanced tumor growth, invasiveness, metastasis, and poor clinical prognosis. CDKN3 also modulates DNA repair, metabolic pathways (such as arginine metabolism), and interacts with immune microenvironment components, contributing to tumor progression and chemoresistance. Its altered expression and subcellular localization serve as biomarkers for various cancers, making CDKN3 a promising therapeutic target and prognostic indicator.
