Overview

Cyclin-dependent kinase-like 4 (CDKL4) is a predicted serine/threonine kinase encoded by the CDKL4 gene, a distant member of the cyclin-dependent kinase family[2]. Its functions are mostly inferred from bioinformatics and genetic association studies. CDKL4 localizes to the cytoplasm and nucleus and may participate in regulating the cell cycle, but its specific substrates and full roles remain largely unvalidated. Clinical and genetic studies suggest that changes in CDKL4 levels are associated with outcomes in colorectal cancer therapy and may impact behavioral responses to acute stress, indicating possible broader effects in tumor biology and neurobiology[2]. Unlike the better-known CDK4, direct drug targeting and functional characterization of CDKL4 are not established. Key caveats: CDKL4 should not be confused with CDK4 ("cyclin-dependent kinase 4"), a therapeutically validated oncology target with approved drugs; CDKL4 is a separate entity with limited experimental validation and is primarily referenced through predictive genetics and association studies rather than direct biochemical data[2]. Current information is incomplete, with much of its biology and druggability still undetermined.
