Overview

Cyclin F (CCNF) is a unique member of the cyclin family distinguished by its F-box domain, enabling its function in the SCF E3 ubiquitin ligase complex. Unlike other cyclins, Cyclin F does not activate cyclin-dependent kinases but instead directly recognizes and mediates the ubiquitination and proteasomal degradation of specific cell cycle and DNA repair proteins, including CP110, RRM2, CDC6, and the E2F family of transcription factors. Its activity is central to proper cell cycle progression, particularly the G2/M transition, and the maintenance of genomic integrity. CCNF is implicated in multiple diseases: loss-of-function mutations contribute to neurodegeneration (ALS/FTD), while dysregulation is associated with cancers such as clear cell renal cell carcinoma, where it serves as a prognostic marker.
