Cyclin I family member 2 (CCNI2) is a member of the cyclin protein family and a homolog of cyclin I (CCNI). It functions as a novel activator of cyclin-dependent kinase 5 (CDK5), differing from other cyclin family activators by localizing CDK5 to the cytoplasm and cell membrane rather than the nucleus. CCNI2 regulates cell cycle progression and cell proliferation, with its depletion leading to decreased cell proliferation and impaired cell cycle progression. Recent research reveals CCNI2 is abnormally elevated in pancreatic cancer and promotes tumor cell proliferation and migration via the PI3K/AKT signaling pathway, highlighting its potential importance as a prognostic marker and as a therapeutic target in cancer. The full physiological and pathological roles of CCNI2 are still being investigated.
As a therapeutic target, inhibition is theorized to impair cell proliferation via interference with PI3K/AKT signaling in cancers expressing high CCNI2
