Overview

Cyclin Y-like protein 1 (CCNYL1) is a member of the **cyclin family**, structurally related to cyclin Y (CCNY), and primarily functions as a regulatory subunit for cell cycle-associated kinases. It binds to cyclin-dependent kinase 16 (CDK16), promoting kinase stabilization and activity. CCNYL1 is highly expressed in the testis, where it is essential for sperm maturation and male fertility; knockout models exhibit profound defects in sperm motility due to disrupted phosphorylation events necessary for proper sperm structure. It also modulates Wnt/β-catenin signaling in mammalian development, with roles in mammary stem cell function and embryogenesis. CCNYL1 acts at the plasma membrane and coordinates various signaling pathways required for neurodevelopment and tissue expansion. There are currently no drugs targeting CCNYL1 directly, nor any evidence for a therapeutically relevant "CCNYL1B" isoform or paralog.
