Cyclo-oxygenase 1 (COX-1), also known as prostaglandin-endoperoxide synthase 1 (PTGS1), is an enzyme that catalyzes the conversion of arachidonic acid to prostaglandin H2 (PGH2), a precursor for various prostanoids including prostaglandins and thromboxanes. COX-1 is constitutively expressed in most tissues and plays a central role in maintaining physiological homeostasis, including gastric protection, platelet aggregation, and renal function. It is a target of nonsteroidal anti-inflammatory drugs (NSAIDs), and its inhibition can lead to gastrointestinal side effects.
