Cyclooxygenase-3 (COX-3) is a variant enzyme related to the cyclooxygenase family, encoded by the PTGS1 (COX1) gene. It differs from COX-1 due to alternative mRNA splicing. While COX-3 may be a functional enzyme in some species, in humans, the translated protein lacks significant cyclooxygenase activity and is unlikely to play any physiological role in prostaglandin synthesis. Acetaminophen may selectively inhibit canine COX-3, but this mechanism is not relevant in humans.
