Overview

CYFIP related Rac1 interactor A (CYRIA/FAM49A) is a cytosolic protein that regulates cellular actin polymerization through direct interaction with **active Rac1** small GTPase[1][2][6][7]. By competing with the WAVE regulatory complex (WRC) for Rac1 binding, CYRIA negatively regulates Rac1 signaling, affecting actin-driven processes such as **cell motility, chemotaxis, cell migration, membrane protrusion formation, and epithelial polarization**[1][7]. CYRIA is structurally homologous to CYFIP proteins (CYFIP1/CYFIP2), sharing the DUF1394 domain, and is highly conserved evolutionarily[1][3]. Altered CYRIA expression has been implicated in cancer progression, particularly promoting cell proliferation and invasiveness when downregulated[1]. CYRIA also contributes to the regulation of immune cell function and host response to bacterial infection by modulating cytoskeletal rearrangement required for pathogen entry[1]. CYRIA has no current direct therapeutic targeting or approved interacting drugs, but may represent a novel candidate for future pharmacological modulation in diseases relating to cytoskeletal remodeling, migration, or infection[2][7][6].
