Overview

CYFIP-related Rac1 interactor B (CYRIB) is a cytosolic protein that regulates the actin cytoskeleton by negatively modulating RAC1 signaling and limiting actin polymerization, particularly by inhibiting the Scar/WAVE complex[1][2][5]. This protein helps control cell migration, chemotaxis, and polarization by focusing protrusive signals and restraining the complexity of pseudopodia. CYRIB plays a role in defending against bacterial entry (notably Salmonella, Listeria, and Mycobacterium), modulates mitochondrial fission and oxidative stress, and is implicated in certain diseases such as cancer, cardiomyopathy, and spinocerebellar ataxia[1][2][5]. No approved drugs are currently known to target CYRIB directly, and its utility as a biomarker or the safety considerations in therapeutic targeting remain undefined in the literature[1][2][5].
