CYMP antisense RNA 1 (CYMP-AS1)

Target
CYMP-AS1
Molecular classification
Long non-coding RNA (lncRNA), Natural antisense transcript
01

Overview

CYMP antisense RNA 1 (CYMP-AS1) is a 2,579-nucleotide long non-coding RNA, mapped to human chromosome 1p13.3. CYMP-AS1 is upregulated in ovarian cancer tissues and localized predominantly in the cytoplasm but also present in the nucleus. It exerts pro-oncogenic functions by interacting with hnRNPM, facilitating its nuclear shuttling, and reducing the stability of AXIN2 mRNA. This regulation promotes activation of the Wnt/β-catenin signaling pathway, advancing cell proliferation, migration, invasion, and EMT. CYMP-AS1 is thus both a mechanistic driver and a novel biomarker for ovarian cancer progression, with proposed targeting strategies including antisense-based modalities.

Mechanism of action

Not applicable; direct-acting drugs are not reported. Mechanistically, targeting CYMP-AS1 (e.g., with antisense oligonucleotides or siRNA) would inhibit its interaction with heterogeneous nuclear ribonucleoprotein M (hnRNPM), restore AXIN2 mRNA stability, and downregulate the Wnt/β-catenin pathway.

03

Biological functions

Regulation of mRNA stability (specifically AXIN2 mRNA)Promotion of cell proliferationPromotion of cell migrationPromotion of cell invasionInduction of epithelial-mesenchymal transition (EMT)Regulation of Wnt/β-catenin signaling pathway
04

Disease associations

Cancer, specifically ovarian cancer (progression, metastasis, prognosis)
05

Safety considerations

No notable safety concerns or therapeutic challenges addressed in the literature. As with other lncRNA-directed therapeutics, off-target effects and efficient delivery are general concerns for future applications.
06

Biomarkers

CYMP-AS1 expression is a biomarker for ovarian cancer progression and poor prognosis, correlating with metastasis and tumor stage
