Overview

CYP3A4 antisense RNA 1 (CYP3A4-AS1; AC069294.1) is a long noncoding RNA located within the CYP3A4 gene locus and transcribed from the antisense strand[1][4]. It acts as a negative regulator of the cytochrome P450 enzymes CYP3A4 and CYP3A5, two major enzymes involved in hepatic drug metabolism. Its expression is inversely correlated with CYP3A4 and CYP3A5 mRNA levels, and it is upregulated by specific genetic variants such as the CYP3A4*1G allele. This upregulation leads to decreased expression of CYP3A4 and CYP3A5, affecting inter-individual variability in drug metabolism and potentially influencing clinical drug responses[1][4]. CYP3A4-AS1 does not itself directly interact with drugs, but it modulates the metabolic capacity of the liver by altering CYP3A4 and CYP3A5 levels. The regulatory mechanism does not appear to extend efficiently to other CYP3A family members (such as CYP3A7 and CYP3A43) under normal physiological conditions[1].
