The cellular cysteine-rich protein (CSRP) family, also known as the CRP family, consists of small LIM-domain proteins including CSRP1, CSRP2, and CSRP3 (Muscle LIM Protein). These proteins are primarily localized in the cytoplasm and nucleus, where they play critical roles in organizing the actin cytoskeleton and regulating gene expression during cell differentiation. CSRP3 is particularly vital in cardiac and skeletal muscle, acting as a mechanical sensor and structural stabilizer within the sarcomere. Dysregulation or mutation of these proteins is strongly linked to human diseases, most notably dilated and hypertrophic cardiomyopathy associated with CSRP3 defects. While they are significant research targets for cardiovascular and oncological therapies, there are currently no FDA-approved drugs that directly target CSRP family members.
