Cysteine and glycine-rich protein 1 (CSRP1) is a LIM domain-containing scaffold protein, widely expressed in mammalian tissues, and highly conserved across vertebrates and invertebrates. CSRP1 functions in gene regulation, cytoskeletal organization, and acts as an integrator in multiprotein complexes governing smooth muscle differentiation, cell migration, and morphogenesis. It regulates expression of genes via interaction with transcription factors like serum response factor (SRF) and GATA proteins; participates in cytoskeletal remodeling via associations with zyxin; and modulates cellular movement and tissue organization via interactions with key signaling pathways such as the noncanonical Wnt and JNK pathways. Aberrant expression or methylation of CSRP1 is implicated in various pathologies including vascular disorders, fibrotic diseases, and cancers such as hepatocellular carcinoma.
