Inhibitors or modulators block the proteolytic activity of ATG4A, disrupting autophagosome formation by preventing ATG8 protein processing and lipidation[3]. Knockdown or inhibition sensitizes cancer cells to chemotherapies by blocking protective autophagy[2].
