Cysteine-rich secretory proteins (CRISPs) are a significant class of non-enzymatic toxins found in the venom of the Eastern Russell's viper, Daboia siamensis (formerly classified as Daboia russelii siamensis). These proteins are characterized by a highly conserved arrangement of sixteen cysteine residues that form eight disulfide bridges, providing a stable molecular framework within the CAP (CRISP, Antigen 5, and PR-1) superfamily (UniProt, 2023). Biologically, CRISPs from this species function primarily as ion channel modulators, specifically targeting voltage-gated calcium channels and cyclic nucleotide-gated channels, which can lead to the inhibition of smooth muscle contraction and disruption of normal physiological signaling (PubMed, 2015). While they are generally less lethal than the phospholipases A2 or snake venom metalloproteinases found in the same venom, they contribute to the overall complexity and synergistic toxicity of the envenomation (Journal of Proteomics, 2015). In a clinical context, these proteins are the targets of antivenom therapy, where specific polyclonal antibodies bind and neutralize the toxins to prevent further systemic damage. Research into these proteins also explores their potential as molecular scaffolds for drug design due to their high specificity for various ion channels (Toxins, 2012).
