Overview

Cysteine-rich secretory protein 2 (CRISP2) is a member of the CRISP family, encoding an intracellular protein highly expressed in the male reproductive tract, specifically within the testis and incorporated during spermatogenesis into the sperm head and tail[1][7]. It functions in the regulation of sperm motility and the acrosome reaction required for oocyte penetration during fertilization[1][7]. CRISP2 is structurally defined by a CAP domain responsible for cell-cell adhesion and steroid binding, and a CRISP domain implicated in ion channel regulation—most notably modulating calcium flux through ryanodine receptors and interacting with the sperm-specific CATSPER calcium channel[1][3]. Decreased or mutated CRISP2 is linked to male infertility, and it exhibits variable expression across mammalian species. In cancer biology, CRISP2 is classified as a cancer-testis antigen, with restricted expression that has prompted investigation as a biomarker, but has not yet been leveraged in targeted therapy or diagnostic applications[2][6].
