Cysteine-rich with EGF-like domain protein 2 (CRELD2) is a multifunctional glycoprotein primarily localized to the endoplasmic reticulum (ER) and Golgi apparatus, possessing protein disulfide isomerase-like activity via multiple CXXC motifs at its N-terminus, which facilitate thiol-disulfide oxidation, reduction, and isomerization essential for ER protein quality control. CRELD2 is inducible under ER stress and can be secreted, particularly in stress conditions, through a process regulated by its C-terminal (R/H)EDL retention motif, and is thought to be involved in cellular responses associated with unfolded protein response, bone/cartilage biology, and homeostasis of various tissues. Its disease associations include cancers, chronic liver, cardiovascular, and kidney diseases, and it may serve as a general marker for perturbed ER homeostasis. It does not currently have known direct drug interactions or established therapeutic interventions.
