Cytidine triphosphate synthase 1 (CTPS1) is an enzyme that catalyzes the conversion of uridine triphosphate (UTP) to cytidine triphosphate (CTP), a critical step in the de novo pyrimidine synthesis pathway. This reaction is essential for the biosynthesis of nucleic acids and phospholipids, making CTPS1 vital for cell proliferation, especially in rapidly dividing cells such as those found in the immune system and certain cancers. Mutations or loss-of-function variants in CTPS1 are linked with immunodeficiency disorders. Overactivity or upregulation has been observed in various cancers, making it an attractive target for anti-cancer therapies.
