Overview

Cytochrome b5 domain-containing protein 2 (CYB5D2), also known as neuferricin, is a secreted, heme-binding protein located mainly at the endoplasmic reticulum membrane. It is part of the membrane-associated progesterone receptor (MAPR) family. CYB5D2 is involved in electron transfer and is essential for regular cellular processes such as inhibition of cell proliferation, induction of apoptosis, and modulation of cytochrome P450 activities involved in steroid synthesis and drug metabolism[2][3]. It functions as a tumor suppressor in various cancers, notably breast and cervical cancer, where downregulation is associated with enhanced tumor growth and poor prognosis[2][5]. CYB5D2 expression also contributes to cell survival during exposure to chemotherapeutic agents, and its loss confers sensitivity to drugs such as paclitaxel, cisplatin, and doxorubicin[2][3]. Mutations affecting its heme-binding domain impair its suppressor functions and its capacity to regulate CYP activities. Additionally, CYB5D2 plays a role in nerve development.
