Cytochrome P450 family 2 subfamily D member 7 (CYP2D7)

Target
CYP2D7
Molecular classification
Enzyme (pseudogene), Cytochrome P450 superfamily, Monooxygenase
01

Overview

Cytochrome P450 family 2 subfamily D member 7 (CYP2D7) is a pseudogene located on chromosome 22 in a cluster with other cytochrome P450 genes, such as CYP2D6 and CYP2D8P. While CYP2D7 and CYP2D6 are highly homologous, CYP2D7 typically contains mutations preventing it from producing a functional protein. Rare functional alleles have been described, but these are extremely uncommon in the population and do not generally produce a catalytically active enzyme in human tissues. CYP2D7 has some relevance in genomics and pharmacogenomics because hybrid alleles and gene conversions between CYP2D7 and CYP2D6 complicate genetic testing and metabolizer phenotype predictions. However, CYP2D7 is not itself a therapeutic target and does not contribute to drug metabolism under normal circumstances

02

Mechanism of action

None (no established biological or pharmacological function)

03

Biological functions

None (pseudogene; major alleles are non-functional)
04

Disease associations

None established (due to non-functionality); rare functional alleles could, in theory, affect drug metabolism, but there is no direct evidence
05

Safety considerations

None specific; hybrid genes with neighboring CYP2D6 can complicate genotype/phenotype predictions for CYP2D6 but do not confer direct activity of CYP2D7 itself
06

Interacting drugs

None (no evidence of drug interaction; see below)
07

Biomarkers

None (no use as biomarker)
